Identification of Drugs Inducing Phospholipidosis by Novel in vitro Data

Drug-induced phospholipidosis (PLD) is a lysosomal storage disorder characterized by the accumulation of phospholipids within the lysosome. This adverse drug effect can occur in various tissues and is suspected to impact cellular viability. Therefore, it is important to test chemical compounds for their potential to induce PLD during the drug design process. PLD has been reported to be a side effect of many commonly used drugs, especially those with cationic amphiphilic properties. To predict drug-induced PLD in silico, we established a high-throughput cell-culture-based method to quantitatively determine the induction of PLD by chemical compounds. Using this assay, we tested 297 drug-like compounds at two different concentrations (2.5muM and 5.0muM). We were able to identify 28 previously unknown PLD-inducing agents. Furthermore, our experimental results enabled the development of a binary classification model to predict PLD-inducing agents based on their molecular properties. This random forest prediction system yields a bootstrapped validated accuracy of 86%. PLD-inducing agents overlap with those that target similar biological processes; a high degree of concordance with PLD-inducing agents was identified for cationic amphiphilic compounds, small molecules that inhibit acid sphingomyelinase, compounds that cross the blood-brain barrier, and compounds that violate Lipinski’s rule of five. Furthermore, we were able to show that PLD-inducing compounds applied in combination additively induce PLD.

We herein report a series of novel rare-earth dialkyl complexes bearing a phosphinoamide anion and demonstrate that the combination of a mono(phosphinoamido)-ligated scandium dialkyl complex with B(C6F5)3 results in an excellent catalyst for the cis-selective cyclization of 1,5-dienes with the ortho-C(sp2)?H bond of pyridines to afford a new family of pyridyl-functionalized 1,3-disubstituted cyclopentane derivatives containing monocyclic, bicyclic, spirocyclic, and heterocyclic skeletons in moderate to excellent yields with high diastereoselectivities (cis/trans up to 99:1).

The mechanism of the gas modifier’s interaction with an analyte in ion separation in drift tube IMS and DMS is explained in this paper.

Orientational ordering in 2,6-lutidine near quartz surfaces modified by carbon

In this paper we present experimental results for the optical dichroism of ultra-thin layers of an epitropic liquid crystal, 2,6-lutidine, on a carbonized quartz substrate. The carbon layer consists of carbon chains with an orientation perpendicular to the substrate surface. It significantly enhances the orientational order of the lutidine layers adjacent to the surface. A two-fold increase of the orientational order parameter S for 2,6-lutidine (from 0.22 to 0.45) has been measured due to the influence of the carbon layer. The inclusion of nitrogen atoms (nN = 7%) into the carbon film leads to a further increase of S from 0.45 to 0.65.

Screening of a library of hemisalen ligands in asymmetric H-transfer: Reduction of aromatic ketones in water

A library of chiral hemisalen ligands (30) was realized. The ligands were synthesized by the condensation of salicylaldehyde derivatives with amino-alcohols (amino-indanol or substituted amino-ethanol) and characterized. These ligands associated with ruthenium (II) precursors were tested on the asymmetric transfer hydrogenation (ATH) of aromatic ketones by sodium formate in water. The different substituent pattern on the ligand (electronic and hindrance effects on different positions) as well as the ruthenium precursor were investigated. The best compromise in terms of conversion and chiral induction led to the complex [RuCl2(mesitylene)]2 coordinated to (1S,2R)-1-((E)-(3-(dimethyl(phenyl)silyl)-2-hydroxy-5-methoxy benzylidene) amino)-2,3-dihydro-1H-inden-2-ol (L25). It reduces acetophenone in 95% yield and 91% ee in 18 h at 30C.

Solid-phase library synthesis of reversed-statine type inhibitors of the malarial aspartyl proteases plasmepsin I and II

With the aim to develop inhibitors of the plasmepsin I and II aspartic proteases of the malaria parasite Plasmodium falciparum, we have synthesized sets of libraries from novel reversed-statine isosteres, using a combination of solution phase and solid phase chemistry. The synthetic strategy furnishes the library compounds in good to high overall yields and with excellent stereochemical control throughout the developed route. The products were evaluated for their plasmepsin I and II inhibiting properties and were found to exhibit modest but promising activity. The best inhibitor exhibits an in vitro activity of 28% inhibition of plasmepsin II at an inhibitor concentration of 0.5 muM (Ki for Plm II=5.4 muM).
