Divergent Strategy for Diastereocontrolled Synthesis of Small- and Medium-Ring Architectures

Nitrogen and oxygen medium rings, in particular nine-membered rings, epitomize a unique area of chemical space that occurs in many natural products and biologically appealing compounds. The scarcity of 8- to 12-membered rings among clinically approved drugs is indicative of the difficulties associated with their synthesis, principally owing to the unfavorable entropy and transannular strain. We report here a scandium triflate-catalyzed reaction that allows for a modular access to a diverse collection of nine-membered ring heterocycles in a one-pot cascade and with complete diastereocontrol. This cascade features an intramolecular addition of an acyl group-derived enol to a alpha,beta-unsaturated carbonyl moiety, leading to N- and O-derived medium-ring systems. Computational studies using the density functional theory support the proposed mechanism. Additionally, a one-pot cascade leading to hexacyclic chromeno[3′,4′:2,3]indolizino[8,7-b]indole architectures, with six fused rings and four contiguous chiral centers, is reported. This novel cascade features many concerted events, including the formation of two azomethine ylides, [3 +2]-cycloaddition, 1,3-sigmatropic rearrangement, Michael addition, and Pictet-Spengler reaction among others. Phenotypic screening of the resulting oxazonine collection identified chemical probes that regulate mitochondrial membrane potential, adenosine 5′-triphosphate contents, and reactive oxygen species levels in hepatoma cells (Hepa1-6), a promising approach for targeting cancer and metabolic disorders.

However, the enantioselective effects of chiral herbicides on nitrogen metabolism have not been comprehensively understood. In this study, effects of chiral herbicide dichlorprop (DCPP) on the nitrogen behaviors in Arabidopsis thaliana were investigated. The results revealed that the uptakes of inorganic nitrogen (NO3- and NH4+) were preferentially impacted by herbicidal active (R)-DCPP, where the transportation of beneficial NO3- was inhibited but the toxic NH4+ was accumulated. As for nitrogen assimilation, enantioselectivity was disappeared and reoccurred when NO3- was assimilated into NH4+. Furthermore, the pathways of NH4+ assimilation differed in Arabidopsis exposed to DCPP enantiomers, where (S)-DCPP remained almost the same to the control relying on GS-GOGAT cycle, but the dominant pathway in (R)-DCPP group was shifted to glutamate dehydrogenase route. Consequently, a profound enantioselectivity was also observed in the effects on amino acids synthesis. As for nitrogen stable isotope fractionation, (R)-DCPP led to more negative delta N-15 values than (S)-DCPP at 0.3 mu M, indicating the selective uptake of toxic NH4+ and different discrimination of delta N-15 was also observed between DCPP enantiomers. Moreover, stable isotopic evidence also revealed the disturbance caused by DCPP to the balance of nitrogen demand and consumption in A. thaliana. All these results may contribute to the final enantioselective toxicity, providing a new sight into the better understanding the action mechanism of chiral herbicides. (C) 2017 Elsevier B.V. Highly Efficient Cyclic Dinucleotide Based Artificial Metalloribozymes for Enantioselective Friedel-Crafts Reactions in Water

The diverse secondary structures of nucleic acids are emerging as attractive chiral scaffolds to construct artificial metalloenzymes (ArMs) for enantioselective catalysis. DNA-based ArMs containing duplex and G-quadruplex scaffolds have been widely investigated, yet RNA-based ArMs are scarce. Here we report that a cyclic dinucleotide of c-di-AMP and Cu2+ ions assemble into an artificial metalloribozyme (c-di-AMP.Cu2+) that enables catalysis of enantioselective Friedel-Crafts reactions in aqueous media with high reactivity and excellent enantioselectivity of up to 97 % ee. The assembly of c-di-AMP.Cu2+ gives rise to a 20-fold rate acceleration compared to Cu2+ ions. Based on various biophysical techniques and density function theory (DFT) calculations, a fine coordination structure of c-di-AMP.Cu2+ metalloribozyme is suggested in which two c-di-AMP form a dimer scaffold and the Cu2+ ion is located in the center of an adenine-adenine plane through binding to two N7 nitrogen atoms and one phosphate oxygen atom.

Comprehensive characterizations including H-1 NMR, FT-IR, UV-Vis, elemental analysis, GPC, and ICP-AES were used to illustrate structures of oligomeric (salen)Mn(III) complexes, while powdered XRD, nitrogen physisorption, together with XPS studies provided further details to detect structures of heterogeneous catalysts. Interestingly, scanning electron microscopy found an interesting morphology change during modification of layered supporting material. Catalytic experiments indicated that configuration of major epoxide products was determined by salen chirality more than that of tartrate linker, but enantioselectivity (e.e. values) could be enhanced when tartrate and salen showed identical chiral configurations. Furthermore, the (R,R)-salen moieties linked with (R,R)-tartrate spacers usually offered higher enantioselectivity compared to other combinations. Lastly, Zn(II)/Al(III) layered double hydroxide played as a rigid supporting material in catalysis, showing positive chiral induction and high recycling potential in catalytic reactions.

The alkaloid (-)-205B has in the past served as a testing ground for novel approaches in nitrogen heterocycle synthesis. We herein report a highly straightforward synthesis of (-)-205B in just six synthetic steps, making it the shortest route currently known. The central steps of our approach are a vinylogous Mukaiyama-Mannich reaction to establish the first two stereogenic centers with excellent diastereo- and enantiocontrol followed by zinc-mediated Barbier allylation to set the third chiral center with high substrate control. Upon cyclization of the Barbier product to lactam and enolate methylation, the third ring is annulated by a one-pot sequence of lactam reduction and aza-Prins cyclization to directly set the final stereogenic center with complete cis-stereoselectivity. As the iminium ion undergoing the aza-Prins cyclization rapidly isomerized to a planar enamine intermediate, the alkaloid was eventually obtained as a 1:1 mixture of C6 diastereomers, which were readily separated by chromatography. Yet, the target natural product was obtained isomerically pure in an overall yield of 12%, which compares favorably with all previous syntheses.
